产品描述：A-443654A-443654 is a potent small-molecule inhibitor of all three Akt serine/threonine kinases, induces Akt Ser-473 phosphorylation in all human cancer cell lines tested, including PTEN- and TSC2-deficient lines._x000D_ IC50 value: 0.16 nM(Ki, Akt1) [1]_x000D_ Target: pan-Akt_x000D_ A-443654 (Ki = 160 pmol/L versus Akt1) inhibits Akt-dependent signal transduction in cells and in vivo in a dose-responsive manner [1]. Rapamycin and A-443654 in combination produced the greatest morphological changes and inhibited cell proliferation by G2/M arrest. Rapamycin and A-443654 in combination induced apoptosis at earlier times and at lower A-443654 concentrations in MCF10CA1a tumor cells than in the benign MCF10A cells [2]. One small molecule, A-443654, showed the greatest selective inhibition of cells with the mutant phenotype. Based on these findings, this inhibitor was screened in vitro against a panel of glioblastoma multiforme cell lines.
